ChIP-seq for RNA Polymerase II with an antibody recognising unmodified CTD and Serine 5 Phosphorylation CTD (clone 4H8). For Each replicate of 100 million ESCs were harvested and cross linked in 1.5mM EGS and 1.5% PFA digested with MNase and mildly sonicated to achieve mono-di-tri nucleosome fragmented chromatin.
