tRNA-seq profiling of Vero E6 cells after SARS-CoV-2 virus infected.Experiments that involved SARS-CoV-2 (Strain CZ02, GenBank MT407650) were conducted in level-3 biosafety laboratories until pure RNA was harvested. Since cells infected with SARS-CoV-2 cannot be centrifugated due to aerosol risks, the final RNA concentration and tRNA-sequencing depth were compromised. Vero E6 cells were cultured in 75 cm2 flasks until ~50% confluence. Then cells were inoculated with SARS-CoV-2 at MOI=0.001 and cultured for ~72 hours until Vero E6 cells reached approximately 25% CPE. After discarding the medium and rinsing twice, cells were lysed and total RNA was extracted and purified using the total RNA extraction kit (Solarbio R1200). Uninfected Vero E6 cells were used as control, and three biological replicates were used for each group.
