Effect of disulfide death related gene ACTN4 on prognosis of lung adenocarcinoma

[Objective] To investigate and screen disulfide death related genes that influence the prognosis and immunotherapy response of lung adenocarcinoma. [Methods] The gene expression profiles and clinical information of normal lung and lung adenocarcinoma samples were downloaded from TCGA. Differential genes were analyzed by Deseq2 package and prognostic correlation was analyzed by Cox regression. The Timer database was used to conduct pan-cancer analysis of target genes, and the patients were divided into high and low expression groups according to the median expression of target genes, and the correlation between target gene expression and clinical features and prognosis was analyzed. Target genes and immune associations were analyzed in TCIA database. Finally, immunohistochemical expression and therapeutic effect of clinical samples were analyzed. [Results] The prognostic disulfide death related genes SLC7A11, CD2AP and ACTN4 were identified, among which ACTN4 had the highest risk ratio. ACTN4 is widely expressed in pancancer species, is significantly reduced in lung adenocarcinoma and can significantly distinguish between the two groups of patients with different prognoses. The expression of ACTN4 was significantly correlated with clinical stage and immune checkpoint expression levels, which further revealed its potential role in lung adenocarcinoma. Immunohistochemical results of clinical samples from our center showed that patients with high expression of ACTN4 in lung adenocarcinoma responded well to immunotherapy. [Conclusion] Disulfide death related gene ACTN4 plays an important role in lung adenocarcinoma, which is expected to provide a certain clinical reference for the treatment of lung adenocarcinoma patients.
