The goal of this study is to investigate the effect of different sugar sources on microglial metabolism. Murine N9 microglia were cultured in T75 flasks under control (standard physiological), high glucose (15 mM) and high fructose (15 mM) conditions for 18 hours. Microglia (250,000/well) were seeded in 24-well plates with the same media as in the T75 flasks and stimulated with LPS (1 µg/ml) for 6 hours. RNA was extracted and gene expression differences between the groups was determined using RNA-seq.
