Whole-genome sequencing was performed on cell-free DNA (cfDNA) isolated from cerebral organoids (COs). H9 embryonic stem cells (ESCs) were used to generate cerebral organoids with the StemDiff Cerebral Organoid Kit and placed in Maturation Medium (StemDiff Cerebral Differentiation Kit; StemCell Technologies) at growth day 10. In addition to day 10 collection, CO samples were collected at time points day 21 and day 41 (maturity). The purpose of the research was determine the feasibility to use CO cfDNA to study and develop biomarkers for neurodevelopmental and neurological disorders.
