Prphp-mCherry inside Grm8

The Prph p -mCherry transgenic mouse model was developed to visualise the expression distribution of a population driven by the Peripherin (Prph) promotor elements and marked by the fluorescence reporter element (mCherry). The integration site of the Prph p -mCherry construct was of interest in the study, as it may have influenced regulatory elements that contributed to the expression distribution of the population in tissue. High molecular weight DNA was extracted from the liver of a C57BL/6J mouse liver (female, 10 weeks), with tape station profiles showing an average fragment size of ~30,000bp after clean-up. These fragments were sequenced with the PromethION platform with a R10.4.1 flow cell, followed by base calling (Raw sequencing data). A de novo assembly located the site of integration (Ravenv1.8.1) and elements of the transgene used to identify the Prph p -mCherry construct. A 2Mbp region of interest (ROI) around the insertion was established based on reads spanning the integration site, which was annotated for establishing features of the integration, for example its copy number and its reading frame (annotated ROI data).
