STR capture is a new technique for generating high-resolution population genomic datasets in species lacking a reference genome. However, STR stutter—the slippage of DNA polymerase on template strands resulting in synthetic new variants—is a persistent obstacle creating genotyping noise. We developed a strategy for stutter correction, and compared traditional and capture STR genotypes in 22 Australian humpback dolphins (Sousa sahulensis) to test the method.NOTE TO USERS: At the time of data release, Sousa sahulensis whole genomic shotgun sequencing (sample 2100M) is undergoing comprehensive analysis and is being released currently to facilitate replicability of the linked publication. We request that users abide by community standards outlined in the Ft. Lauderdale and Toronto Agreements, and refrain from publishing genome-wide analyses using these data until the data producing research group has done so first.
