Soil was collected from the Buck pasture plot at the Hopland Research and Extension Center (Mendocino County, CA) at a depth of 0–10cm on 10/03/2020. Soil was sieved through a 2.0mm wire mesh. Microcosms were established by adding 206g (+/- 0.5g) of soil to 64oz acid washed and autoclaved mason jars. Each microcosm had one of the following treatments: (1) natural abundance water (diH2O); (2) diH2O + KH2PO4; (3) heavy water (H218O); (4) H218O + KH2PO4 in triplicate (four treatments x three replicates x three timepoints = 36 mason jars). H218O enrichment ranged from 98.06–98.54% and KH2PO4 (for +P treatments) was added to the water and mixed for a final concentration of 50µM. Water was added to each sample slowly and evenly across the soil surface with a syringe to bring the soil up to 30% moisture. Mason jars were sealed with lids that had airtight septa and incubated at room temperature (~23°C).
This dataset represents sequencing of the v4 16S and 5.8S/ITS4 ITS regions of all but 8 samples. Missing samples include: T_2_H218O_P_A, T_3_H2O_A, T_3_H2O_B, T_3_H2O_C, T_3_H2O_P_A, T_3_H2O_P_B, T_3_H2O_P_C, and T_3_H218O_A. Duplicate T0 dry soil was sequenced and 1 water control for a total of 31 samples. Each sample was seqeunced for 16S and ITS for a total of 62 amplicon samples. Each amplicon had paired reads for a total of 124 reads.
