Cell proliferation during starvation in Aiptasia (Fig. S5H)

After 60min EdU pulse (300uM, 2%DMSO), aposymbiotic or symbiotic polyps of <i>Aiptasia</i> CC7 were dissociated using ACME. Incorporated EdU was labeled by Click-it reaction with Alexa488 fluorophore and cells were stained with 1μg/ml FxCycle violet DNA dye (Invitrogen). Flow cytometry was performed a BD LSRFortessa (BD Life Sciences) and the resulting data was analysed using FlowJoV10.8 (BD Life Sciences).. EdU-Alexa488 fluorescence in aposymbiotic and symbiotic animals was analysed separately across several starvation time points (1day - 2day - 5day - 12day - 21day) and based on a comparison with DMSO controls, the EdU index was recorded. .wsp files contain the samples, gating strategy and cell populations used for the analysis in FlowJo.fcs files represent the individual flow cytometry output files that were analysed in the .wsp file: <br>
