The pathogenesis of endometritis in dairy cows is related to pathogenic microorganisms, local inflammatory injury and uterine microecological disorders. Escherichia coli (E. coli) is the main causative pathogen of endometritis in dairy cows. Therefore, we investigated E. coli-induced endometritis mechanisms in dairy cows using bovine bone marrow-derived macrophages (BMDMs).E. coli infection was performed on BMDMs for 4 hours. BMDMs were pretreated with PGD2 inhibitors (H-PGDS Inhibitor 1, HQL-79) for 24 hours, followed by E. coli infection for 4 hours. Samples were collected and evaluated. Four types of samples were collected: uninfected macrophages, macrophages infected with E. coli for 4 hours, and macrophages pretreated with a PGD2 inhibitor (H-PGDS Inhibitor 1, HQL-79) for 24 hours before being infected with E. coli for 4 hours. Each sample type was replicated six times. First, we compared uninfected samples with E. coli-infected samples to identify genes pertinent to the study. Next, we compared the PGD2 inhibitor-treated samples with the E. coli-treated samples.
