Roles of Staphylococcus aureus alternative sigma factor B in staphylococcal bovine mastitis

A sigB null (?sigB) mutant was constructed and analyzed for its phenotypes and transcriptome along with those of the parental RF122 strain. Method: Duplicate samples of rRNA depleted RNA from wild type and mutants were used to study transcriptomes by ion torrent platform. Overall design: rRNA-depleted RNA samples from stationary phase wilde type and sigB null mutant cultures were used to compare transcriptomes. Some affected genes from RNAseq result were selected for confirmation by quantitative reverse transcriptase PCR.
