Early mammalian erythropoiesis requires the DOT1L histone methyltransferase. Single-cell suspensions prepared from E10.5 Dot1L- methyl mutant, Dot1L-knockout and wildtype mouse yolk sacs were cultured for 3 days in StemPro34 media supplemented with nutrient supplement, 2U/ml human recombinant EPO, 100ng/ml SCF, 10uM dexamethasone , 40ng/ml IGF-1 and penicillin-streptomycin. Total RNA was purified from these cultured cells, libraries were prepared, and RNA-sequencing was done to identify Dot1L-regulated genes.
