Huh6 and Hep-G2 were co-cultured with macrophages directly. NCI-H23 was co-cultured with conditional medium of macrophages and SPP1(osteopontin). Cells were seeded into 6-well plate (200,000 cells per well), and then human monocyte-derived macrophages were added into each well (100,000 cells) for 24 hours. NCI-H23 cells were stimulated with 30% of conditional medium of macrophages or 10ng/mL SPP1(osteopontin) for 24 hours. Based culture madium was DMEM/HamF12 medium supplemented with 10% FBS.
