High throughput direct RNA sequencing of MOLM13 leukaemia cell line to study the interplay of post transcriptional modifications after METTL3 knockdown.

Direct RNA sequencing enables the simultaneous assessment of RNA modifications, gene expression, polyadenylation dynamics and alternative splicing. Here, we use the direct RNA sequencing approach to sequence the MOLM13 leukaemia cell line on the high throughput PromethION instrument to generate sufficient yield and coverage for the study of the interplay of m6A RNA modifications and other post transcriptional modifications when the m6A writer METTL3 is knocked down.
