Pbr1 Allele Alignment

Alignment of Pbr1 allele sequences (genomic DNA of start codon to stop codon) from 12 barley genotypes, aligned by Clustal Omega. Allele sequences were obtained by Sanger sequencing PCR fragments. This alignment was used to generate the phylogenetic tree in Figure 5C in Carter and Helm, et al 2018 (in submission).<br>From methods section:DNA was isolated from ground frozen leaf tissue using the GeneJET Plant Genomic DNA Purification Kit (Thermo Scientific™). Primers were designed throughout the genes of interest and fragments were amplified from genomic DNA using Q5 2X Master Mix (NEB), then Sanger sequenced at the Cornell Biotechnology Resource Center.For the analysis of <i>Pbr1</i> alleles, nucleotide sequences were selected from each sequenced allele that spanned from the start codon to the stop codon of the Rasmusson allele, including the intron. Sequences were aligned with Clustal Omega.<br>
