We placed immotile variants of the soil bacterium Pseudomonas fluorescens SBW25 (SBW25(delta)fleQ IS-(omega)Km-hah: PFLU2552) under selection for recovered flagella-mediated motility using soft agar as a directional selection medium. We observed that typically a single nucleotide polymorphism (SNP) would arise de novo within 72-120 hours and restore the motility phenotype. Throughout this experiment we identified a single rare mutant line that was a double mutant, harbouring a single deletion in the gene glnK (glnK del.84) and a single nucleotide polymorphism in the RNA polymerase sigma factor 54 (rpoN A688C). This secondary mutation was identified through the whole genome sequencing.
