Isolation of grassland soil bacteria on different corrinoid conditions

By studying corrinoids (the vitamin B12 family of cofactors) as model metabolites, we aim to study complex metabolic interactions among soil microbes mechanistically. The purpose of this study was to determine whether corrinoid type impacts the culturability of soil bacteria. We isolated bacteria from a California grassland soil on adapted VL60 media amended with one of six corrinoids or no corrinoid. We identified these isolates by 16S rRNA amplicon sequencing and sequenced the genomes of a subset, in addition to experimental characterization of their metabolism.
