As a new source of natural fibres, the Bombax ceiba tree can provide the thinnest, lightest and an extremely soft warm fibre material for the textile industry. As we know, the natural fibre cell initiation process not only determines fibre yields but is also an ideal model for cell growth and differentiation studies. However, the molecular mechanisms underlying fibre initiation regulation remain unclear. In this study, we found that fibre cells were differentiated from the epidermis of the inner wall in the ovary of B. ceiba. Then, each initiated cell divides into a cluster of fibre cells and eventually develops into mature fibres, which is very different from the classical fibre initiation process in cotton. A high-throughput single-cell RNA sequencing platform was used to describe the characterization of the fibre cell initiation process in B. ceiba. A total of 347 potential marker genes for fibre initiation cell types were screened out. By comparing the gene clusters involved in the fibre cell initiation process between B. ceiba and cotton, we filtered candidate genes and key regulators that may contribute to fibre cell differentiation and division in B. ceiba. Overall, our study revealed expression signatures during fibre initiation in B. ceiba at single-cell resolution, and this provides a new strategy and viewpoint for the investigation of natural fibre cell differentiation and development.
