Successful derivation of mouse trophoblast stem cells (TSCs) and trophectoderm stem cells (TESCs) from blastocysts greatly facilitates understanding of trophoblast differentiation. However, the understanding of early trophectoderm differentiation remains incomplete. We report a stable morula trophectoderm stem cells (MTSCs) derived from 32-cell embryos that show enhanced and uniform trophoblast core gene expression. Meanwhile, MTSCs can self-organize to form organoids, and the differentiated organoids contain almost all placental cell types.
