The gut is a vital organ in the human body, and gut fungi play a crucial role in maintaining intestinal health. However, research on gut fungi remains limited compared to other areas of gut microbiota. In this study, we identified a novel fungal strain, from the gut microbiota that exhibits significant therapeutic potential for repairing intestinal inflammation. Further investigation revealed that Pt produces a bioactive compound, DC12, which can be purified and demonstrated robust efficacy in alleviating intestinal inflammation. Mechanistic studies showed that DC12 binds to the enzyme enolase 1 (ENO1) and subsequently promotes the expression of Yes-associated protein 1 (YAP1). This interaction facilitates the repair and regeneration of intestinal epithelial cells, thereby accelerating intestinal recovery. Our findings highlight the therapeutic potential of gut fungi-derived compounds and provide a novel strategy for treating intestinal inflammatory disorders.
