To identify CHIP mutation frequency in myeloid cells in blood and in cardiovascular tissues; To compare gene expression profiles of blood or cardiovascular tissue myeloid cells from CHIP mutation carriers and non-carriers. Overall design: Blood myeloid cells, blood lymphocytes, myeloid cells from human carotid plaque or heart tissue were isolated using fluorescence-activated cell sorting(FACS) in bulk. DNA and RNA of the same sorted cells were isolated. The DNA were used for quantifying the size of CHIP mutated clones of each cell type acquired from the CHIP mutation carriers. The RNA of myeloid cells from carriers and matched non-carriers were subjected to RNA-seq for gene expression profile comparison.
