To search for enzymes in the secoiridoids biosynthesis pathway, we collected fruits of six commercial olive cultivars with different oleuropein content for sequencing: the sweet varieties Dolce d'Andria (DAN) and Tendellone (TEN), suitable to produce table olives because of low oleuropein levels, as well as four varieties containing medium to high levels of phenolic secoiridoids, Arbequina (ARB), Leccino (LEC), Coratina (COR) and Moraiolo (MOR), Olives from were harvested from collection fields of CNR IBBR in Perugia (Italy) every 20 days from 45 (stage 1) until 125 (stage 5) days after flowering. Olive fruit mesocarp was frozen in liquid nitrogen, pulverized using a mortar and pestle, and stored at – 80 °C until needed. RNA was extracted from frozen and milled olive fruit mesocarp using Plant RNeasy® Kits (QIAGEN; Hilden, Germany) and sent for RNA-Sequencing to BGI Genomics (Shenzhen, China) following the company's protocols.
