Cappable-Seq (Ettwiller et. al., BMC Genomics volume 17, Article number: 199 (2016)) was employed to identify transcriptional start sites (TSSs) representing the primary transcriptome of hormogonia, motile filaments of the filamentous cyanobacterium in N. punctiforme ATCC29133. TSSs were defined using the wild-type strain and subsequently the expression of these TSSs was analyzed in each of three sigma-factor deletion mutant strains. Total RNA from a single biological replicate at 6 hours after hormogonium induction from either the wild-type strain, or sigJ (UOP132), sigC (UOP131), and sigF (UOP141) mutant strains (Gonzalez et. al., mSphere 4(3) 2019) was used to prepare cDNA libraries for Cappable-Seq as described by (Ettwiller et. al., BMC Genomics volume 17, Article number: 199 (2016)), apended with adapters and single-read sequenced in 1 lane of a flow cell on an Illumina NextSeq 500 system using 75 bp read length
