This project investigates transcriptional changes in aortic macrophages derived from Tet2-deficient hematopoietic stem cells in a mouse model of Angiotensin II induced aortic injury. ApoE knockout mice were lethally irradiated and reconstituted with bone marrow cells from Tet2 wild type or Tet2 knockout donor mice. After one month of engraftment, Angiotensin II was infused for three days and the aorta was collected. Aortic macrophages were isolated by FACS sorting based on CD45, CD64, Ly6G negative, and Ly6C negative gating. Bulk RNA sequencing was performed to characterize gene expression programs associated with Tet2 deficiency in aortic macrophages.
