Effect and mechanism of silencing EIF3E gene on glioma cell line (second part)

<b>Objective:</b><b> </b>The aim of this study was to investigate the effect and mechanism of EIF3E gene silencing on human glioblastoma T98G cell line.<b>Methods:</b><b> </b>A silenced EIF3E cell model was constructed in T98G cells, and transcriptome data affected by silenced EIF3E gene were obtained using high-throughput transcriptome sequencing, and corresponding gene changes were analyzed.<b>Results: </b>Silencing of the EIF3E gene in T98G cells caused a significant upregulation of the expression levels of 460 genes, which were enriched in the top ten biological pathways, including type I interferon signaling pathway, negative regulation of viral genome replication, response to virus, defense response to virus, innate immune response, immune system process, response to beta interferon, response to alpha interferon, nucleosome assembly, and interferon-mediated. Therefore, it is speculated that EIF3E may play a regulatory role in the immune system of human glioblastoma by regulating the expression of these genes. A total of 1562 genes were found to have significantly downregulated expression levels, and these EIF3E negatively regulated genes were enriched in the top ten biological pathways, including ciliary assembly, protein transport, cell cycle, cell division, non-motile cilium assembly, positive regulation of GTPase activity, cellular projection organization, docking of basal body to membrane, intraflagellar transport in cilia assembly, and centrosome duplication.<b>Conclusio</b><b>n:</b><b> </b>1. EIF3E may regulate a large number of genes in human glioblastoma, especially immune-related genes. At the same time, it has a preference for regulating gene expression, and the number of downregulated genes after EIF3E silencing is far greater than that of upregulated genes, suggesting that the EIF3E gene may have a function of stabilizing gene expression. 2. The expression of transcription factors changed after EIF3E silencing, suggesting that EIF3E may interact with transcription factors to regulate gene expression.
