To systematically study the biological role of microRNA-­‐200s in pancreatic beta cells we conditionally inactivated the miR-­‐200a/200b/429-­‐allele in a constitutively miR-­‐141/200c-­‐deficient background. These mice are viable, normal weight and normoglycemic indicating no gross metabolic alterations under basal conditions. We are currently studying the pathophysiological functions of miR-­‐200 using different models of beta cell dysfunction including diet-­‐induced insulin resistance. To understand the underlying mechanisms of miR-­‐200 functions we will perform RNA-seq analysis to identify target genes of miR-­‐200. The overall aim of the study is to identify the molecular mechanisms underlying miR-­‐200 mediated effects in metabolic disease.
