To systematically study the biological role of microRNA-‐200s in pancreatic beta cells we conditionally inactivated the miR-‐200a/200b/429-‐allele in a constitutively miR-‐141/200c-‐deficient background. These mice are viable, normal weight and normoglycemic indicating no gross metabolic alterations under basal conditions. We are currently studying the pathophysiological functions of miR-‐200 using different models of beta cell dysfunction including diet-‐induced insulin resistance. To understand the underlying mechanisms of miR-‐200 functions we will perform RNA-seq analysis to identify target genes of miR-‐200. The overall aim of the study is to identify the molecular mechanisms underlying miR-‐200 mediated effects in metabolic disease.
