Astrocytes become activated in a mouse model of microglial ablation. We isolated astrocytes from hippocampus after microglial ablation, and performed RNA-seq analyses to reveal gene expression profile of activated astrocytes. Hippocampus was dissected from two Siglechdtr/dtr mice 2 days after PBS or DT injection, and astrocytes were isolated using magnetic-activated cell sorting system with anti-astrocyte cell surface antigen-2 MicroBeads (Miltenyi Biotec). Three pools of RNA were obtained from three independent experiments. Libraries were prepared using the SMARTer Stranded RNA-seq Kit (Clontech Laboratories) followed by sequencing with NovaSeq6000 (Illumina).
