This study aimed to determine what genes are differentially expressed following 2 hours' treatment with napabucasin (BBI-608) alone or in combination with NQO1 depletion. RNA-sequencing of MiaPaCa2 cells ("MiaPaca2 Parental," cells purchased from ATCC), MiaPaCa2 cells stably expressing either the Rosa26 control vector ("MiaPaCa2 Rosa26") or MiaPaCa2 cells stably expressing different CRISPR guide-RNAs to mutate the NQO1 gene, resulting NQO1 loss ("MiaPaCa2 NQO1-71" and "MiaPaCa2 NQO1-163)", treated for 2 hours with 500 nM napabucasin or with DMSO as vehicle control. For each cell line and treatment condition, 3 biological replicates were performed, except for MiaPaCa2 Rosa26 which has 2 biological replicates for the vehicle-treated control.
