We allowed the rhizosphere bacterium Pseudomonas protegens CHA0 to evolve on the roots of Arabidopsis thaliana for six plant growth cycles. To quantify changes in bacterial phenotypes, sixteen evolved bacterial colonies were randomly selected from each of the five plant replicate selection lines at the end of the second, fourth and sixth growth cycle, in addition to sixteen randomly selected ancestral colonies (in total 256 isolates). Then k-means analysis was applied to determining novel bacterial phenotypes. A subset of isolates of each bacterial phenotypes was characterised for their effects on plant root architecture and above and belowground biomass in separate assays at the end of the selection experiment.
我们使根际细菌产蛋白假单胞菌(Pseudomonas protegens)CHA0在拟南芥(Arabidopsis thaliana)的根部完成六轮植物生长周期的演化。为量化细菌表型的变化,我们在第二、第四及第六轮生长周期结束时,从5个植物重复选择系中各随机选取16株演化后的细菌菌落,同时额外随机选取16株祖先菌落,总计获得256株细菌分离株。随后采用K均值(k-means)分析鉴定新型细菌表型。在演化选择实验结束时,我们选取对应每种细菌表型的部分分离株,通过独立测定实验表征其对植物根系构型以及地上、地下生物量的影响。
