To investigate the effect of intracellular signals mediated by chemokine receptor 5 (CCR5) on osteoclast differentiation, bone marrow cells were harved from Ccr5-deficient mice (KO) and their littermates (WT) mice. Bone marrow cells were harvested from tibia of 8 weeks-old male Ccr5-deficient mice and WT. Cells were induced into bone marrow macrophages with M-CSF for 2 days, and further differentiated into osteoclast precursors (pOCs) with RANKL for 2 days. pOCs were then subjected to RNA-sequencing.
