Transcriptomic profiling granulomatous fractons in C3HeB/FeJ mice infected with M. tuberculosis

Necrotic granulomatous lesions within the lungs of C3HeB/FeJ mice infected with M. tuberculosis were microdissected into three fractions, Caseum, foamy macrophages, and Cell. RNA were extracted from fractions, followed by rRNA depletion with using NEBNext rRNA Depletion Kit (NEB). Libraries were constructed with NEBNext Ultra II Directional RNA Library Prep Kit (NEB). Libraries were examined with Qubit fluorometer (Thermo Fisher Scientific) and GenNext NGS Library Quantification Kit (TOYBO) for quantity control, and DNA High Sensitivity Kit on Agilent Bioanalyzer 2100 (Agilent) for quality control, respectively. Paired-end sequencing (75 bp x 2) of libraries were performed using NextSeq500 (Illumina).
