Stanniocalcin-2 (STC2), a glycosylated protein discovered in the endocrine glands of fish, has been shown to play multiple cell biological roles in cancers. However, the function and specific mechanism of STC2 in colorectal cancer (CRC) remain unclear. Herein, bioinformatics analysis and CRC tissue specimens were used to investigated the expression and prognostic value of STC2. Flow cytometry, sphere formation and Dead/Live staining assay were used to examine the biological functions of STC2, and the specific mechanism was explored using RNA sequencing, RNA immunoprecipitation (RIP) assays and RNA stability assay. In this study, we found that STC2 was upregulated in CRC, and its highly expression was correlated with poor prognosis in CRC patients. STC2 could promote resistance to anoikis in CRC, which through upregulating TGIF1 expression. Mechanistically, we found that STC2 regulated TGIF1 by binding to the mRNA of TGIF1 which repressing its degradation. Our study revealed that STC2 could be a potential therapeutic target for CRC treatment and provide potential new insights into the strategy of CRC treatment.
