microCT dataset of Amphiporus sanakensis n.sp. (SIO-BIC W10103) - HOLOTYPE
收藏资源简介:
The anterior part of the holotype of Amphiporus sanakensis n.sp. from Sanak Seep in the NE Pacific (SIO-BIC W10103) was scanned using a Werth Tomoscope® XS Plus 200 microCT (Giessen, Germany). The specimen had been fixed in formalin and subsequently preserved in 70% ethanol. Before scanning, it was stained for 3 weeks with a 0.3% phosphotungstic acid (PTA) solution in 70% ethanol for contrast enhancement. The scanning parameters were as follows: SIO-BIC W10103 was scanned using a voltage of 80 kV, a generator current of 140 µA, and a generator power of 11.2 W. The exposure time was set at 666 ms across a total of 1500 projections, with 4 projections averaged to enhance image quality. This yielded a voxel resolution of 8.81 µm. Surface renderings of the specimens were created using Drishti v3.1 software (Australian National University, Canberra).
