A global transcriptional analysis of representative high (UM6)and low (UM1) clinical B. pseudomallei biofilm producers was performed to identify the genes required for biofilm formation in B. pseudomallei. We utilized RNA sequencing and comparative transcriptome analysis to identify genes and their respective expression levels that contribute to the B. pseudomallei biofilm phenotype of both UM6 and UM1. Total RNA was isolated from two biological replicates of B. pseudomallei UM6 and UM1 harvested at early logarithmic growth phase. After removal of rRNA, cDNA libraries were constructed from the RNA samples and sequenced on an Illumina HiSeq2000 at up to 8 million fragments per sample.
