GROQ-seq Function Measurements for T7 RNA Polymerase SSVL and epPCR Libraries.

This dataset contains GROQ-seq functional measurements of T7 RNA polymerase variant libraries — a site-saturation variant library (SSVL) and an error-prone PCR (epPCR) library, covering 34,000 variants across >12,000 single- and 21,000 multi-residue mutants. Fitness is measured by linking T7 RNAP activity to bacterial growth via mDHFR-mediated trimethoprim resistance, using transcriptional output at a T7 promoter as the function readout. This run of the assay was performed at the Design, Automation, Manufacturing, and Processes (DAMP) Laboratory at Boston University. Part of a broader effort to generate sequence → function data for polymerases, supporting protein-engineering applications in synthetic biology, mRNA therapeutics, and biosensors.
