Identification of NAT10 targets in CRC by RNA immunoprecipitation and acethyl-RNA immunoprecipitation

To identify potential mRNAs that are acetylated by NAT10, RIP with a NAT10 antibody and acRIP with a ac4C antibody were conducted in DLD-1 and SW480 cells Total RNA isolated from DLD-1 or SW480 cells was framented and subjected to immunoprecipitation using anti-ac4C antibody. After incuation and washing, samples were eluted using elution buffer containing ac4C nucleoside. The resultant final product was used for RNA-seq. Besides, RNA immunoprecipitation was performed in DLD-1 and SW480 cells according to manufacturer's protocol. RNA isolated from RIP was also used for RNA-seq.
