This study investigates the impact of splenectomy on the cardiac immune cell landscape post-I/R injury. We performed single-cell RNA sequencing (scRNA-seq) on CD45+ immune cells isolated from mouse hearts 24 hours after I/R injury in both splenectomized and sham-operated (control) mice. Overall design: Male C57BL/6 mice aged 8 weeks were randomly assigned to two groups: splenectomy and sham surgery. both groups underwent myocardial ischemia-reperfusion (I/R) injury induced by occluding the left anterior descending (LAD) coronary artery for 60 minutes, followed by 24 hours of reperfusion. Hearts were then harvested, and CD45? immune cells were isolated using magnetic-activated cell sorting (MACS).
