The Zn-hypersensitive EMS mutant ozs3 (overly zinc sensitive) shows some additional pleiotropic effects compared to Col-0 wildtype. Most severe effects are a complete growth arrest at 4°C (seedling stage) and an altered root morphology even under control conditions. To detect the underlying transcriptional differences we analyzed the root transcriptome under control and cold stress (4°C, 24h) conditions. Plants were cultivated for four weeks in a hydroponic system under short day conditions (8h light, 16h dark, at 23 °C). For treatment plants were transfered to 4°C for 24 h under short day conditions (8h light, 16h dark). Control plants remained at 23°C (8h light, 16h dark). After treatment roots of control- and cold-stressed-plants were harvested and total RNA was extracted using RNeasy Plant Mini Kit from Qiagen. Finally Arabidopsis Genome ATH1 Arrays (Affymetrix) were hybridized. Three biological replicates were analyzed for each condition and genotype.
