We generated a barcoded vector library using a PCR primer containing random bases, ligated it to a mispaired DNA library, and transformed the resulting plasmid library into E. coli. By comparing the relative abundance of repaired and unrepaired molecules, we directly quantified the relative repair efficiency of many different mismatches in multiple sequence contexts in parallel.
