Environmental-(e)DNA approaches have the potential to provide unprecedented insight into the structure of marine benthic communities at a fraction of the cost of morphological-based (i.e. microscope) methods. In a proof of concept study, matching molecular and morphological data sets were created from six benthic samples collected from sediments of the Western English Channel Observatory site L4. We also aimed to determine best practice for sample collection and compared DNA extraction from 10 g of sediment with 5 mL of filtered sediment porewater (i.e. free DNA present outside any organism). For all samples, both the 16S and 18S rRNA genes were sequenced using the Earthmicrobiome V4 and V9 PCR primes, respectively.
