Computational redesign of a thermostable T7 RNA polymerase

This Zenodo repository contains all data and novel code used for the paper "Computational redesign of a thermostable T7 RNA polymerase" by authors Zachary T. Baumer and Tim A. Whitehead. This repository contains:(1.) SOURCE_DATA_FILE.xlsx contains all experimental data presented in the paper. This file also contains all primers, plasmid sequences, and gBlock sequences used for plasmid construction. (2.) The Jupyter notebook ‘Evaluating Tmapp from CD.ipynb’ is a custom script for extracting Tm,app from circular dichroism temperature melts. (3.) PROSS_outputs.xlsx lists the full PROSS outputs and our relative ranking of the mutations for testing in this paper.
