This dataset provides a controlled benchmark for evaluating batch correction methods in flow cytometry analysis. The dataset consists of splenic cell samples from C57BL/6 mice with synthetically introduced technical batch effects, designed to enable rigorous quantitative assessment of batch normalization algorithms. Dataset Design: Biological samples: Splenic cells from 3 wild-type C57BL/6 mice (2 female, 1 male) plus 2 mutant strains.Technical replicates: Each biological sample split into 3 technical replicatesSynthetic batch effects: Created by systematically varying antibody concentrations across three staining panelsMarkers: 12-color panel including CD3, CD25, CD8, CD44, CD62L, CD44, IgD, IgM, NK1.1, Ly6C, B220, CD4, viability dye.
