The biology of S. pombe Pus1

Schizosaccharomyces pombe pus1 mutants are temperature sensitive due to reduced levels of tRNAIle(UAU), as a consequence of the lack of pseudouridylation at residues 27, 34, and 36. To understand mechanisms by which the pus1 mutants can overcome or prevent the loss of tRNAIle(UAU), we isolated spontaneous suppressors of the temperature sensitivity, and then did whole genome sequencing of the suppressors to identify the mutations in the chromosome that were responsible for the suppression.
