The Kalash are an isolated population of Indo-European speakers who have been living for centuries in the Hindu Kush mountain ranges of northern Afghanistan and Pakistan and experienced strong bottle-neck and genetic drift during the course of its history. To obtain insights into their origins and demography, we propose to sequence one individual at high coverage (~30x). We have chosen a male carrying Y haplogroup L3a because this Y lineage is of particular additional interest: haplogroup L3a is present at a high frequency (23%) in the Kalash but has not been observed in any other population from Pakistan, and L3a has not been included in other sequencing studies that we know of. The DNA has been prepared from a lymphoblastoid cell line that was established for the Human Genome Diversity Project.
