Labcode 1- HIV neutralization testing with pseudovirions

This protocol includes a detailed protocol of HIV neutralizaion assay based on pseudovirus produced by co-transfection of pEnv with a lentiviral expression plasmid, pGJ3-luc, into HEK293T cells. The HEK293T cells was used in a neutralisation assay with monoclonal antibodies and sCD4. Target cells were 3T3 cells engineered to express CD4 and CCR5 or CXCR4, and the readout measured luciferase reporter gene expression.
