Given the link between commensal microbiota and Th17 development, there is intense interest in understanding the role of IL-17 in regulating host defense genes expression in the gut. To Assess this we performed RNA sequencing on terminal ileum of the following mouse strains WT, Il17a-/-, Il17ra-/-, and intestinal epithelial specific IL-17RA knockout mice.To understand the role of IL-17 in regulating host defense gene expression in the intestinal epithelial cells. Towards this we cultured primary intestinal enteroids from C57BL/6 mice and stimulated with recombinant IL-17A for 24 hours. We performed RNA sequencing on recombinant IL-17A (50 ng/ml) stimulated enteroids.
