Single-cell sequencing performed on endothelial cells after exposure to focused ultrasound in vivo to determine the transcriptomic signature of the response to intervention. Overall design: Focused Ultrasound Blood-Brain Barrier Opening was performed in eGFP-Cldn5 mice targeting the striatum and putamen. The animals were sacrificed after 1 or 72 hours and the targeted brain tissue was dissected out and prepared for FACS. The cells were sorted based on eGFP expression to isolate endothelial cells using FACS, and the cells were submitted for single cell RNA sequencing.
