We designed and synthesized 23 pairs of primers in the entire exons, and 3 kb of 5′ and 3′ flanking regions of bovine LIPJ gene according to the genomic sequences of UMD3.1.1 (GenBank accession number: AC_000183.1). 44 semen DNAs were pooled to use for the PCR amplifications with the equal concentration (50 ng/µL) for each DNA. All the PCR products were sequenced by ABI3730XL DNA analyzer (Applied Biosystems, Foster, CA, USA). Then, we aligned the sequences with the reference genome (UMD3.1.1) using the BLAST (https://blast.ncbi.nlm.nih.gov/Blast.cgi), and identified the novel SNP, g.10250120A/G.
