Dendritic cells (DCs) simultaneously prime and dampen the immune response. The transcriptional program that regulates immunosuppression in DCs is currently unknown. Here we identified ETS homologous factor as a key transcription factor that regulates DC immunosuppression. Mice were immunized intraperitoneally (i.p.) with or without 10 µgTLR stimulation in 100 µL PBS, peritoneal cavity cells were obtained by peritoneal cavity lavage twice with 5 mL of ice-cold PBS. They were collected 6 hrs after injection for cDCs (live MHCII+ CD11c+) FACS-sorting for scRNA-seq library preparation.
