Genomic changes in tumor cell lines can occur during culture, leading to differences between cell lines carrying the same name. Because authentication based on short tandem repeat analysis identifies only the cell line of origin and disregards genetic drift, characterisation of cell lines works for reference genome profiles and contributes to reproducibility of the results. The Ishikawa cell line originated from a well differentiated human endometrial adenocarcinoma and has been used for more than 35 years following its establishment in 1980 (Nishida et al., 1996). In this project, we emply chromosome sorting to generate chromosome-specific DNA and perform chromosome sequencing to identify breakpoint junctions and distinguish variants between homologues.
